Target Information
CHEK1 (CHK1) is a serine/threonine protein kinase involved in preventing replication when DNA integrity is compromised.
CHK1 is a key mediator in the DNA damage-induced checkpoint network and an evolutionarily conserved kinase ensuring genomic integrity under genotoxic stress.
Inhibition of CHK1 abrogates the G2 or S checkpoint, leading p53-deficient cancer cells to mitotic catastrophe and apoptosis, while normal cells remain arrested in G1.
Thus, CHK1 inhibitors can enhance the efficacy of DNA-damaging agents in cancer cells, making Chk1 an attractive therapeutic target, especially since about 50% of human cancers are p53-deficient.
