Target Information
Amphiregulin (AR) binds to the EGF receptor (EGFR) with lower affinity than EGF. The mature secreted AR is an 84-amino acid glycosylated growth regulator derived from a 252-amino acid transmembrane precursor. Seven distinct ligands, including EGF, TGFα, AR, HB-EGF, cripto-1, epiregulin, and betacellulin, can bind to the extracellular domain of EGFR. These ligands share a characteristic EGF-like domain defined by six cysteine residues forming three disulfide-bonded loops. AR protein is localized in epithelial tissues such as colon, stomach, pancreas, breast, and placenta, and is overexpressed in several human cancers.
