Target Information
Amphiregulin (AR) binds to the EGF receptor (EGFR) with lower affinity than EGF. The mature secreted form of AR is an 84-amino acid glycosylated polypeptide growth regulator, generated by proteolytic processing of a 252-amino acid transmembrane precursor.
Seven distinct polypeptide ligands—EGF, TGFα, AR, HB-EGF, cripto-1, epiregulin, and betacellulin—bind to the extracellular domain of EGFR.
All these growth factors share an EGF-like domain defined by six evenly spaced cysteine residues forming three loops via disulfide bonds.
AR protein is localized in the epithelium of the colon, stomach, pancreas, breast, and placenta, and is reportedly overexpressed in human cancers of breast, colon, stomach, and pancreas.
