Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage and activated mature T and B lymphocytes.
CD25 binds IL-2 with low affinity but forms a high-affinity IL-2 receptor complex with CD122 (IL-2Rβ) and CD132 (common γ chain). The PC61.5 antibody inhibits IL-2 binding to both receptor classes, playing a role in lymphocyte differentiation and activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal results.

Fluorophore Information:
PE-eFluor 610 can be excited with 488–561 nm laser lines and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red equivalent). Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

CD25 (IL2 receptor alpha chain, IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. IL2RA forms a high-affinity receptor complex with IL2RB and IL2RG.
IL2 acts as a growth and differentiation factor, regulating cell death and immune responses. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency and immune dysfunction.
