Target Information
PIF1 is a single-stranded (ss) DNA-dependent ATPase and DNA helicase that unwinds DNA in a 5′ to 3′ direction relative to its binding strand. It plays a critical role in DNA replication and telomere length maintenance in Saccharomyces cerevisiae.
The PIF1 gene is highly conserved from yeast to humans. Mutations in PIF1 lead to telomere lengthening, indicating its function as a catalytic inhibitor of telomere formation and elongation.
Human PIF1 preferentially binds telomeric DNA and inhibits telomerase activity. It counteracts RecQ homolog Sgs1 helicase activity, and its degradation is mediated by the ubiquitin–26S proteasome pathway.
