Target Information

The tachykinins are a family of small peptides including neurotransmitters such as substance P, neurokinin A, and neurokinin B. These act on three distinct seven-transmembrane G protein-coupled receptors at varying concentrations.
The NK-3 receptor (NK3R) shows the highest affinity for neurokinin B and is highly expressed in the supraoptic and paraventricular nuclei. Upon ligand binding, NK3R activates a phosphatidylinositol-calcium second messenger system, likely triggering the release of vasopressin and oxytocin. NK3R may also play roles in learning and memory, as knockout mice show cognitive deficits. Although NK3R has been suggested to regulate vagal afferent relay neurons, these receptors are more likely activated by substance P or neurokinin A since airway nerves do not express neurokinin B.
