Target Information
Estrogen Receptor beta (ERβ, NR3A2) is a nuclear receptor protein of approximately 55 kDa, with isoforms ranging from 36–59 kDa. Initially cloned from human testis, ERβ belongs to the steroid/thyroid hormone receptor superfamily. It functions as a ligand-activated transcription factor that binds estrogen and interacts with estrogen response elements (EREs) in DNA to regulate gene transcription.
Two forms of estrogen receptor exist — alpha (ESR1) and beta (ESR2). ERβ binds estrogens with affinity comparable to ERα, and certain isoforms inhibit ERα activity in reproductive tissues. ERβ is widely expressed and acts as a tumor suppressor, with roles in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular risk regulation.
