TRAM2은 collagen type I 합성에 필수적인 단백질로, ER Ca²⁺ pump인 SERCA2B와 translocon의 활성을 연결하여 콜라겐 합성 부위에서의 국소 Ca²⁺ 농도를 증가시킬 수 있습니다. 이 높은 Ca²⁺ 농도는 콜라겐 폴딩에 관여하는 분자 샤페론의 기능에 필요합니다. 또한 TM4SF20의 첫 번째 막관통 나선 N-말단의 ER 내강 삽입에 필요하며, 세라마이드 감지자로 작용하여 조절된 대체 전이(RAT)에 관여할 수 있습니다.
