IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine and a member of the IFN–IL-10 family (including IL-19, IL-20, IL-24, IL-26, IL-28, IL-29, and type I/II interferons).
Produced mainly by activated T cells and NK cells, IL-22 gene is located on chromosome 12q and structurally related to IL-10. It binds to a heterodimeric receptor complex composed of IL-22R1 and IL-10R2, activating the JAK–STAT signaling pathway.
A soluble IL-22 receptor (IL-22BP) antagonizes IL-22 binding and signaling. IL-22 primarily targets nonimmune cells such as skin, digestive, respiratory cells, hepatocytes, and keratinocytes.
It acts as an effector cytokine of the Th17 lineage, co-expressed with IL-17A and IL-17F, regulating genes associated with innate immunity in the skin. IL-22 contributes to host defense against extracellular pathogens, wound healing, and inflammation through activation of STAT3.
