Target Information

STAT3는 STAT (signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동 후 사이토카인 및 성장인자 자극에 반응하여 전사인자로 작용합니다. CREB-binding protein과 같은 공동활성화 인자가 STAT3의 전사 활성화에 필요합니다.
STAT3는 Interferon-alpha, Interferon-gamma, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, Tyr705 위치의 인산화는 JAK1 및 JAK2에 의해 매개되어 STAT3 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다.
STAT3 유전자는 인간 17번 염색체 q arm에 위치하며, 다양한 세포 자극에 대한 유전자 발현을 매개하여 세포 성장 및 세포사멸 등 여러 세포 과정에 관여합니다. Rac1 GTPase는 STAT3의 활성을 조절하고, PIAS3 단백질은 STAT3의 특이적 억제인자입니다.
STAT3 결손 생쥐는 초기 배아 단계에서 치명적인 것으로 보고되었습니다.
