Target Information
Caspases는 세포 사멸(apoptosis)의 주요 매개체 역할을 하는 cysteine protease 계열입니다. 모든 caspase는 prodomain과 큰/작은 촉매 서브유닛으로 구성된 전구체 형태로 존재하며, 활성형은 다양한 자극(리간드-수용체 상호작용, 성장인자 결핍 등)에 의해 생성됩니다. Caspase-3는 Yama, CPP32, apopain으로도 알려져 있으며, 32 kDa 단백질을 암호화합니다. Caspase-3는 apoptosis 동안 PARP를 85 kDa 형태로 절단하며, CrmA 단백질에 의해 억제될 수 있습니다. 추가 기질에는 DNA-PK, actin, GAS2, procaspase-6 등이 포함됩니다. 활성화는 Asp-28/Ser-29 및 Asp-175/Ser-176 부위 절단을 통해 17 kDa 및 12 kDa 서브유닛을 형성하여 a2b2 테트라머로 활성 효소를 구성합니다.
