Product Specific Information

Recombinant human-mouse chimeric monoclonal antibodies are produced using in vitro expression systems. Recombinant antibodies are generated from specific genes encoding the desired antibody, cloned into expression vectors and expressed in vitro.
Advantages include improved specificity and consistency between lots.
It is recommended to titrate the antibody carefully for optimal performance in the assay of interest.

Target Information

This antibody recognizes the constant region of the T cell receptor (TR) alpha chain (PubMed:24600447).
Alpha-beta T cell receptors are antigen-specific receptors essential for immune response, located on the surface of T lymphocytes. They recognize peptide-major histocompatibility (pMH) complexes presented by antigen-presenting cells, a key step for adaptive T cell immunity (PubMed:25493333).
Binding of alpha-beta TR to pMH complexes triggers TR-CD3 clustering and LCK activation, leading to phosphorylation of CD3 subunits and recruitment of ZAP70. ZAP70 phosphorylates LAT, forming the LAT signalosome that activates calcium, MAPK, and NF-κB pathways, promoting transcription factor mobilization critical for T cell growth and differentiation (PubMed:9382891, PubMed:23524462).
The T cell repertoire is generated in the thymus via V-(D)-J rearrangement and shaped by selection events to produce self-MH restricted, non-autoaggressive T cells. Post-thymic interactions further refine TR structural and functional avidity (PubMed:15040585). [UniProt]
