Target Information
IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine in the IFN-IL-10 family (including IL-19, IL-20, IL-24, IL-26, IL-28, IL-29, and type I/II interferons).
Produced mainly by activated T cells and NK cells, IL-22 gene is located on chromosome 12q and structurally related to IL-10.
It acts through the heterodimeric receptor complex IL-22R1 and IL-10R2, triggering JAK-STAT signaling.
A soluble receptor, IL-22BP, antagonizes IL-22 binding and signaling.
IL-22 primarily targets nonimmune cells such as skin, digestive, respiratory cells, hepatocytes, and keratinocytes.
It is an effector cytokine of Th17 lineage, co-expressed with IL-17A and IL-17F, regulating innate immunity genes.
Functions include induction of acute phase reactants, antimicrobial proteins, and enhancing keratinocyte mobility.
Highly expressed during chronic inflammation, IL-22 activates intracellular kinases and transcription factors, crucial for host defense and wound healing.
It mediates IL-23-induced acanthosis and dermal inflammation via STAT3 activation.
