The monoclonal antibody 5E3 reacts with human and mouse Semaphorin 4A (Sema4A), a 150 kDa glycoprotein of the class IV semaphorin family that exists in membrane-bound and soluble forms. Sema4A is expressed in lung, kidney, spleen, testis, mammary glands, and activated T cells and dendritic cells. It binds to Tim-2, neuropilin-1 (Nrp1), plexin D1, and plexin B1, and plays roles in T cell activation, Th1 differentiation, and Treg function and survival.

Testing shows that 5E3 antibody stains resting CD49b+ NK cells in mouse but not CD56+ NK cells in human.

• Applications Tested: Pre-titrated and tested for flow cytometry of normal human peripheral blood cells
  • Recommended use: 5 µL (0.06 µg) per test
  • A test = staining a cell sample in 100 µL final volume (10⁵–10⁸ cells/test)

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm, excited by 488 nm blue laser. Use a 710/50 bandpass filter (695/40 acceptable alternative). Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and samples from light.

Samples can be stored in:

• 1-step Fix/Lyse Solution (Product #00-5333-57)
  Store up to 3 days at 4°C in the dark with minimal signal loss.
