Description:
The GIGD7 monoclonal antibody reacts with mouse TIGIT (T cell immunoreceptor with Ig and ITIM domains, also known as VSTM3 or WUCAM), a 26 kDa member of the poliovirus receptor (PVR) family. In mice, TIGIT is expressed on follicular T helper cells. In humans, it is expressed on NK cells, regulatory T cells, follicular T helper cells, memory CD4⁺ and CD8⁺ T cells, but not on B cells or naïve CD4⁺ T cells. Expression is upregulated upon T cell activation.

TIGIT interacts with PVR and PVR-like family members such as CD155 and mediates interactions between NK/T cells and antigen-presenting cells, fibroblasts, and endothelial cells expressing PVR proteins.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤1 µg/test. Adjust cell number empirically (10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). It can substitute for PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be fixed using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  Store up to 3 days at 4°C in the dark with minimal effect on brightness and FRET compensation. Clone-specific performance should be verified empirically.

Target Information

TIGIT binds with high affinity to the poliovirus receptor (PVR), leading to increased IL10 secretion and decreased IL12B secretion, thereby suppressing T cell activation by promoting the generation of mature immunoregulatory dendritic cells.
