Product Specific Information
The V9 monoclonal antibody recognizes human Vimentin, a 57 kDa protein that functions as a structural component of intermediate filaments. Vimentin is expressed in cells derived from the mesenchyme and specific populations such as radial glia, immature glial cells, and pancreatic precursor cells. It is also proposed as a marker of cardiac differentiation. In neural cells, vimentin expression is gradually replaced by neurofilaments. Reports indicate surface expression on activated macrophages, platelets, apoptotic T cells, and neutrophils.
This antibody also recognizes canine, rat, and chicken vimentin but does not recognize mouse vimentin.
Applications Reported: Immunohistochemical staining of frozen and paraffin-embedded tissue sections, and immunocytochemistry.
Applications Tested: Tested by immunocytochemistry of fixed and permeabilized cells at ≤5 µg/mL. Optimal titration recommended.
eFluor® 660 is a replacement for Alexa Fluor® 647, emitting at 659 nm and excited with the red laser (633 nm).

Target Information
Vimentin is a developmentally regulated intermediate filament protein (IFP) found in cells of mesenchymal origin. It is involved in intracellular protein transport between the nucleus and plasma membrane. During development, vimentin is replaced by tissue-specific IFPs. It plays a role in steroid synthesis by acting as a storage network for cholesterol-containing lipid droplets. Phosphorylation of vimentin leads to filament breakdown and cytoskeletal rearrangements facilitating cholesterol transport to mitochondria, increasing steroid synthesis.
