This Mgc3 monoclonal antibody detects hypoxia-inducible factor 1 alpha (HIF-1α) from human, non-human primate, bovine, mouse, and porcine cells. It does not cross-react with ARNT or HIF-2α.

• Validated Applications: Western blotting, immunofluorescence, immunocytochemistry, immunoprecipitation, flow cytometry, and gel shift assays.
• Western Blot: Detects ~116 kDa HIF-1α after hypoxic induction in COS cells.
• Immunofluorescence: Shows nuclear staining in COS-7 cells after 1% O₂ exposure for 4 hours.
• Flow Cytometry: Tested on DFOA-stimulated HeLa cells (methanol fixation). Recommended ≤1.0 µg/test.
  • A test = antibody amount for 100 µL cell sample (10⁵–10⁸ cells/test).
  • Optimize titration for best results.

Target Information

HIF1-alpha (HIF1A) is a subunit of the HIF1 transcription factor, which regulates cellular responses to hypoxia. HIF1 is a heterodimer composed of alpha and beta subunits, both belonging to the basic-helix-loop-helix PAS family.

• Function: Activates transcription of genes involved in oxygen delivery and metabolic adaptation under hypoxia (e.g., EPO, GLUTs, VEGF).
• Regulation: Interacts with p53 and MDM2 under hypoxia; promotes degradation via ubiquitination.
• Pathway Involvement: Apoptosis, cell proliferation, angiogenesis, ROS regulation.
• Chromosomal Location: Chromosome 14q (human).
• Binding Partners: C-terminal binds p300; p300/CBP-HIF complexes induce hypoxia-responsive genes.
• Clinical Relevance: Plays a role in ischemia, cancer, pulmonary hypertension, and chronic obstructive pulmonary disease.
