Target Information

The CD40 antigen is a single-chain glycoprotein belonging to the tumor necrosis factor/nerve growth factor superfamily. It shows significant homology to the Hodgkin’s disease-associated antigen CD30. CD40 is expressed on all B cells except plasma cells, and also on certain epithelial cells, carcinomas, and lymphoid dendritic cells.

CD40 plays a crucial role in mediating immune and inflammatory responses such as:

• T cell-dependent immunoglobulin class switching
• Memory B cell development
• Germinal center formation

AT-hook transcription factor AKNA regulates CD40 expression, contributing to homotypic cell interactions. Adaptor protein TNFR2 interacts with CD40, mediating signal transduction. The CD40-CD40L interaction is involved in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms of CD40 have been reported.
