Target Information

The low density lipoprotein receptor (LDL-R) gene family consists of cell surface proteins involved in receptor-mediated endocytosis of specific ligands. LDL normally binds at the cell membrane and is internalized into lysosomes, where the protein is degraded and cholesterol becomes available for repression of HMG CoA reductase, the rate-limiting enzyme in cholesterol synthesis. Concurrently, cholesterol ester synthesis is stimulated.
Mutations in the LDL-R gene cause familial hypercholesterolemia, an autosomal dominant disorder. LDL-R, along with SCARB1, CLDN1, and CD81, is also implicated as an entry factor for the Hepatitis C virus. At least three isoforms of LDL-R are known.
