Target Information
NALP proteins include the apoptosis regulator APAF1 and mammalian NOD-LRR proteins, which are thought to be involved in inflammation and reproduction.
NALP4 (PYRIN-containing APAF1-like protein 4) possesses a C-terminal leucine-rich repeat (LRR) region, an N-terminal Pyrin domain (PYD), a NACHT domain, and a NACHT-associated domain.
In transfected 293 cells, NALP4 suppressed NF-kappa-B induction by TNF-α and IL-1-β, indicating that NALP4 acts at a convergence point in these signaling pathways. It also inhibited NF-kappa-B activation triggered by overexpression of TRAF2, TRAF6, RIP, IRAK2, IKK-α, and IKK-β, suggesting a critical role in modulating NF-kappa-B activity.
