Product Specific Information

Description:
The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein and a member of the selectin family of adhesion receptors. It includes:

• N-terminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes, including lymphocytes, monocytes, and polymorphonuclear cells, except for a subset of memory T cells.
