Product Specific Information

eBioscience offers a panel of monoclonal antibodies to different epitopes of human Foxp3, providing tools for investigating Foxp3 expression and subsets of Foxp3⁺ cells.
The PCH101 antibody reacts with the amino terminus of human Foxp3 protein (FORKHEAD BOX P3, SCURFIN, JM2). Cross-reactivity to other proteins has not been determined.

Foxp3 (49–55 kDa) is a transcriptional regulator defective in scurfy mice. High Foxp3 mRNA expression occurs in CD4⁺CD25⁺ regulatory T cells (Treg), and ectopic expression in CD4⁺CD25⁻ cells induces a Treg phenotype.

Intracellular staining of human PBMCs using the anti-human Foxp3 Staining Set reveals 0.5–4% lymphocyte staining, mainly in CD25^bright population (subject to donor variability).
PCH101 cross-reacts with rhesus, chimpanzee, and cynomolgus. Recommended companion antibodies include CD4 (OKT4 or RPA-T4) and CD25 (BC96).

Applications Reported: Intracellular staining followed by flow cytometry.
Applications Tested: Pre-titrated and tested for intracellular staining and flow cytometry using Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523).
Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.
Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis.
It is expressed at high levels in CD25⁺CD4⁺ regulatory T cells, low in CD4⁺/CD25⁻ cells, and absent in CD4⁻/CD8⁺ T cells.
FOXP3 defects cause scurfy phenotype in mice and IPEX syndrome in humans.
Transcript variants encoding different isoforms have been identified. In breast and colon cancer cells, FOXP3 expression is regulated by p53 in response to DNA damage.
