The G-protein-coupled receptor (GPCR) superfamily includes approximately 600–1,000 members and represents the largest class of molecular targets with therapeutic relevance.
The nociceptin receptor (also known as the orphanin FQ receptor) is a G-protein coupled opioid receptor that binds the endogenous neuropeptide nociceptin. Ligand binding induces conformational changes that trigger signaling via G proteins, modulating downstream effectors such as adenylate cyclase and calcium channels.
Arrestins regulate G protein signaling and activate alternative pathways leading to MAP kinase activation. The receptor plays a key role in nociception, pain perception, and regulation of locomotor activity.
