Target Information

Quinidine is an alkaloid extracted from the bark of the Cinchona tree with class 1A antiarrhythmic and antimalarial effects.
It stabilizes neuronal membranes by binding to and inhibiting voltage-gated sodium channels, reducing sodium influx required for impulse initiation and conduction.
This increases the excitation threshold and decreases depolarization during phase 0 of the action potential.
Additionally, the effective refractory period (ERP), action potential duration (APD), and ERP/APD ratios are increased, resulting in decreased conduction velocity.
Quinidine’s antimalarial activity arises from its action as an intra-erythrocytic schizonticide, binding to heme polymer (hemazoin) in the parasite’s acidic food vacuole and preventing further polymerization, leading to toxic heme accumulation and parasite death.
