Store vial at -20°C prior to opening. Aliquot contents and freeze at -20°C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

IL-27 / p28 antibody is purified by a multi-step process including delipidation, salt fractionation, and ion exchange chromatography followed by extensive dialysis against the buffer stated above. This antibody is specific for mouse and rat p28 protein. Cross-reactivity with IL-27 from other sources has not been determined.

Target Information

IL-27, a member of the IL-12 family, is a heterodimeric protein consisting of the p40-related protein Epstein-Barr virus-induced gene 3 (EBI3) non-covalently linked to an IL-12p35-related protein, p28 (also known as IL-30). IL-27 is produced by activated antigen-presenting cells (APCs) and mature dendritic cells.

IL-27 acts on NK cells and naive CD4+ T cells, and receptor expression analysis (WSX-1/TCCR and gp130) suggests it may also affect mast cells and monocytes. Binding of IL-27 to WSX-1/gp130 activates JAK1, STAT1, and STAT3, leading to their phosphorylation.

WSX-1/TCCR-deficient mice show impaired Th1 responses and increased susceptibility to L. monocytogenes infection, indicating IL-27’s essential role in initiating Th1 responses. IL-27 induces the Th1 transcription factor T-bet and suppresses the Th2 factor GATA-3, supporting Th1 differentiation.

Recent studies also show IL-27 has potent antitumor activity by enhancing CTL function, inhibiting tumor growth, and inducing regression of neuroblastoma tumors.
