Target Information
MCL1 (Myeloid cell leukemia-1)은 Bcl-2 패밀리에 속하며 세포 생존 유지에 관여합니다. Isoform 1은 세포사멸(apoptosis)을 억제하고, Isoform 2는 이를 촉진합니다. MCL1의 발현은 DNA 손상 요인(이온화 방사선, 자외선, 알킬화제 등)에 노출된 ML-1 세포에서 증가하며, 이는 GADD45와 Bax 증가 및 bcl-2 감소와 연관됩니다.
MCL1은 미토콘드리아에 주로 존재하며, bcl-2와 함께 세포의 생존을 촉진합니다. 그 작용 기전은 명확하지 않으나, Bcl-2 패밀리의 pro-apoptotic 단백질(Bim, Bak 등)과 이합체를 형성해 기능을 억제하는 것으로 알려져 있습니다.
MCL1은 다양한 세포 유형에서 발현되며, 배아 발생 및 B, T 림프구 발달에 필수적입니다. 두 가지 주요 isoform(MCL1L, MCL1S)이 존재하며, MCL1S의 과발현은 세포사멸을 유도합니다.
