Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDKs) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and acts as a major transcriptional target of p53. In the nucleus, p21 binds and inhibits CDK1 and CDK2, blocking the transition from G1 to S phase or from G2 to M phase after DNA damage, allowing DNA repair. In the cytoplasm, p21 exhibits anti-apoptotic activity by binding to caspase 3 and apoptotic kinases ASK1 and JNK. p21 has a dual role in carcinogenesis, functioning as both a tumor suppressor and an oncogene depending on context.
