Target Information

HMCES acts as a sensor of abasic sites in single-stranded DNA (ssDNA), preserving genome integrity by promoting error-free repair of abasic sites. It binds abasic sites at replication forks and forms a covalent cross-link with DNA via a thiazolidine linkage between the lesion and its catalytic cysteine residue. The resulting DNA-protein cross-link is degraded by the proteasome.

This mechanism protects abasic sites from error-prone polymerases and endonucleases that could cause mutations or double-strand breaks. HMCES also participates in class switch recombination (CSR) in B-cells by binding ssDNA overhangs and promoting DNA double-strand break repair through the microhomology-mediated alternative-end-joining (Alt-EJ) pathway.

Additionally, HMCES mediates autocatalytic processing of its N-terminal methionine to expose the catalytic cysteine.
