Target Information
Spleen tyrosine kinase (SYK) is a cytoplasmic tyrosine kinase involved in immune receptor signaling. Along with ZAP70, it is considered a potential drug target for allergic disorders, autoimmune diseases, and certain cancers. Upon activation of B cell antigen receptors or high-affinity IgE receptors, Syk translocates to the plasma membrane and phosphorylates adaptor proteins, initiating downstream signaling pathways related to calcium mobilization, cytoskeletal remodeling, and gene transcription. Syk interacts with receptor ITAM motifs via its SH2 domains and is further activated by Src-family kinase phosphorylation.
