Target Information
Upon entry into mitosis, the cell’s microtubule (MT) network forms the mitotic spindle, enabling segregation of paired chromosomes. Proteinaceous structures on centromeric chromatin, termed kinetochores (KT), are essential for proper attachment of chromosomes to spindle MTs.
A spindle and kinetochore complex composed of SKA1, SKA2, and SKA3 proteins is required for stable KT–MT interactions and timely anaphase onset. Depletion of SKA3 by siRNA delays anaphase transition, causing a prolonged metaphase-like state. These SKA3-depleted cells accumulate high levels of checkpoint protein Bub1 at kinetochores, indicating that the SKA complex is crucial for spindle checkpoint silencing and maintenance of chromosome cohesion during mitosis.
