Product Specific Information
PA1-962 detects proteasome 19S subunit 10B from canine, hamster, human, mouse, and rat tissues and cells.
It has been successfully used in Western blot procedures.
By Western blot, this antibody detects ~42, 46, 48, and 52 kDa proteins corresponding to subunit 10B and possibly combined signals from subunit proteins 4, 6B, 7, and 8.
The immunizing peptide corresponds to amino acid residues 278–294 of human proteasome 19S subunit 10B.
This sequence is conserved in 19S protein subunits S4 (~53 kDa), S6A (~53 kDa), S6B (~46 kDa), S7 (~47 kDa), and S8 (~45 kDa).
PA1-962 immunizing peptide (Cat. # PEP-107) is available for use in neutralization and control experiments.
Target Information
The 26S proteasome is a multicatalytic proteinase complex composed of two complexes: a 20S core and a 19S regulator.
- The 20S core consists of four rings of 28 non-identical subunits: two rings of seven alpha subunits and two rings of seven beta subunits.
- The 19S regulator includes a base (6 ATPase subunits and 2 non-ATPase subunits) and a lid (up to 10 non-ATPase subunits).
Proteasomes are abundant in eukaryotic cells and cleave peptides in an ATP/ubiquitin-dependent, non-lysosomal pathway.
The immunoproteasome plays a key role in processing class I MHC peptides.
This gene encodes one of the non-ATPase subunits of the 19S regulator lid and may also participate in TNF signaling via interaction with the TNF type 1 receptor.
A pseudogene has been identified on chromosome 1.
