CD40 항원은 종양괴사인자/신경성장인자 수용체 초가족에 속하는 단일 사슬 당단백질로, Hodgkin 병 관련 항원 CD30과 높은 상동성을 보입니다.
CD40은 대부분의 B 세포(형질세포 제외), 일부 상피세포, 암세포 및 림프성 수지상세포에서 발현됩니다.
이 단백질은 T 세포 의존적 면역글로불린 클래스 전환, 메모리 B 세포 발달, 생식 중심 형성 등 다양한 면역 및 염증 반응에 필수적입니다.
AT-hook 전사 인자 AKNA는 CD40 발현을 조절하며, TNFR2 어댑터 단백질은 신호 전달 매개체로 작용합니다.
CD40과 그 리간드의 상호작용은 아밀로이드-베타 유도 미세아교세포 활성화에 필수적이며, 알츠하이머병 병태생리의 초기 단계에서 중요한 역할을 합니다.
CD40의 두 가지 대체 스플라이싱 변이체가 보고되었으며, 관련 질환으로는 Type 3 Hyper-IgM 면역결핍증 및 CD40 리간드 결핍증이 있습니다.
