Background
This gene encodes a potent cyclin-dependent kinase inhibitor. The encoded protein binds to and inhibits the activity of cyclin-CDK2 or cyclin-CDK4 complexes, regulating cell cycle progression at the G1 phase. Its expression is tightly controlled by the tumor suppressor protein p53, mediating p53-dependent G1 arrest in response to stress stimuli. The protein also interacts with proliferating cell nuclear antigen (PCNA), playing a regulatory role in S phase DNA replication and DNA damage repair. It can be cleaved by CASP3-like caspases, leading to activation of CDK2 and contributing to apoptosis execution.
