Mature rat IL-2 was expressed in Pichia pastoris X-33 (GenBank Accession # P04351, amino acids 21–155).
The expressed protein was N-terminal sequenced and begins with the following 10 amino acids: APTSSPAKET.
Purity is >95% by SDS-PAGE analysis.

Activity:
Biological activity was determined by measuring proliferation of mouse CTLL-2 cells.
ED50 (1 unit of activity) ≈ 3 ng/mL.

Use:
For in vitro applications. Perform dose-response assay to determine optimal concentration for specific experiments.

Reconstitution and Dilution:

• Store lyophilized material at 4°C.
• Reconstitute in 0.5 mL distilled or deionized water.
• Dilute in buffer containing carrier protein, aliquot, and store ≤ -70°C.
• Avoid multiple freeze-thaw cycles.
• Suitable buffers: PBS with 1% BSA or neutral media containing <10% FCS.

Expression of IL-2 in mature thymocytes is monoallelic, representing a unique regulatory mechanism.
Disruption of IL-2-like genes in mice leads to ulcerative colitis-like disease, highlighting its role in immune response regulation.
