Target Information
Caspase 8 (CASP8)은 cysteine-aspartic acid protease (caspase) 계열에 속하는 단백질로, 세포 사멸(apoptosis)의 실행 단계에서 중심적인 역할을 합니다. Caspases는 비활성 전구체 형태로 존재하며, 내부 aspartic residue에서의 절단을 통해 활성화되어 큰 및 작은 서브유닛으로 구성된 효소를 형성합니다.
Caspase 8은 Fas 및 다양한 apoptotic 자극에 의해 유도되는 프로그램된 세포 사멸에 관여하며, FADD와의 상호작용을 통해 신호 전달에 참여합니다. 또한 Huntington 병 환자의 뇌 조직에서 Caspase 8이 검출되어 신경퇴행성 질환과의 연관성이 제시되었습니다.
활성화된 Caspase 8은 Caspase 9 등 하위 caspase를 활성화하여 세포 사멸을 유도합니다. Jurkat 세포 및 편도 조직에서도 반응하며, 과발현 시 apoptosis를 유도하지만 ICE family 특이적 억제제에 의해 차단될 수 있습니다. 다양한 splice variant가 보고되었으나, 모든 isoform의 full-length 서열은 규명되지 않았습니다.
