Target Information
FOXO1 (FKHR, Forkhead Box O1)는 forkhead homeotic 유전자군에 속하는 전사인자입니다. FOXO1은 핵 수용체 활성 조절에 관여하며, PAX3와의 결합은 alveolar rhabdomyosarcoma와 관련이 있습니다. 또한 FOXO1은 자가포식, 근육 성장 및 분화, 산화 스트레스에 대한 대사 항상성 조절 등 다양한 세포 기능에 핵심적인 역할을 합니다.
FOXO1은 insulin response element (IRE, 5'-TT[G/A]TTTTG-3') 및 Daf-16 family binding element (DBE, 5'-TT[G/A]TTTAC-3')에 결합하며, 골세포 수 조절 및 골량 유지, 골격계의 내분비 기능을 통한 포도당 대사 조절에도 관여합니다.
간세포에서 FOXO1은 PPARGC1A와 함께 작용하여 IGFBP1, G6PC, PCK1 유전자 발현을 활성화함으로써 포도당신생합성을 촉진합니다. 또한 FOXO1은 CDK1, PKB/AKT1, SKT4/MST1 경로 하류에서 세포 사멸을 조절하는 중요한 인자입니다.
