The G-protein-coupled receptor (GPCR) superfamily consists of approximately 600–1,000 members and represents the largest known class of molecular targets with proven therapeutic value.
The nociceptin receptor is a G-protein coupled opioid receptor that functions as a receptor for the endogenous neuropeptide nociceptin. Ligand binding induces a conformational change that triggers signaling via G proteins, modulating downstream effectors.
This signaling mediates inhibition of adenylate cyclase and calcium channel activity. Arrestins modulate G protein signaling and activate alternative pathways, including MAP kinase activation.
The receptor plays a role in modulating nociception and pain perception and contributes to the regulation of locomotor activity by nociceptin.
