Target Information
The UBE2V1–UBE2N and UBE2V2–UBE2N heterodimers catalyze the synthesis of non-canonical Lys-63-linked polyubiquitin chains, which do not lead to proteasomal degradation. These heterodimers mediate transcriptional activation, regulate cell cycle progression, differentiation, and DNA repair pathways.
They cooperate with E3 ligases such as HLTF, SHPRH, RNF5, RNF135, and TRIM5 to promote Lys-63-linked polyubiquitination of substrates including PCNA, JKAMP, RIG-I/DDX58, and TRAF6, activating NF-κB and MAPK signaling and contributing to antiviral and inflammatory responses.
