0.2 µg/mL of MA5-45213 was sufficient for detection of HSP70 in 20 µg of heat-shocked HeLa cell lysate by colorimetric immunoblot analysis using Goat anti-mouse IgM:HRP as the secondary antibody. Detects approximately 70 kDa. May detect HSP70, HSC70, and HSP72.

Target Information

HSP70 is a heat shock protein expressed in response to various biological stresses, including high temperatures. The HSP70 family is highly conserved and induced by stress in all examined organisms. Human HSP70 family members include:

• HSP70: Inducible and constitutively expressed in primate cells
• HSP72: Induced exclusively under stress conditions
• HSC70: Constitutively expressed, involved in clathrin-coated vesicle uncoating
• GRP78 (BiP): Glucose-regulated 78 kDa protein localized in the endoplasmic reticulum
• HSP75 (p75): 75 kDa mitochondrial protein

HSP70 is encoded by an intronless gene and, together with other heat shock proteins, stabilizes existing proteins against aggregation and assists in folding newly synthesized proteins. It also participates in the ubiquitin-proteasome pathway via interaction with AU-rich element RNA-binding protein 1. The HSP70 gene is located in the MHC class III region, clustered with two related genes encoding similar proteins.
