1 µg/mL of MA5-45210 was sufficient for detection of HSP70 in 20 µg of heat-shocked HeLa cell lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody. Detects approximately 70 kDa. May detect HSP70, HSC70, Grp78, and HSP72.
Target Information
HSP70 is a heat shock protein expressed in response to various biological stresses, including high temperature. The HSP70 family includes highly conserved proteins induced by stress in all organisms.
Major human members include:
- HSP70: inducible in all organisms, constitutive in primate cells
- HSP72: induced exclusively under stress conditions
- HSC70: constitutively expressed, involved in clathrin-coated vesicle uncoating
- GRP78 (BiP): ER-localized, glucose-regulated protein
- HSP75 (p75): mitochondrial protein
HSP70 stabilizes existing proteins against aggregation and assists in folding newly synthesized proteins. It also participates in the ubiquitin-proteasome pathway via interaction with AU-rich element RNA-binding protein 1. The HSP70 gene is located in the MHC class III region, clustered with related genes encoding similar proteins.
