LDLRAD4 functions as a negative regulator of TGF-beta signaling and may play roles in:

In the canonical TGF-beta pathway, ZFYVE9/SARA recruits SMAD2 and SMAD3 to the receptor. After phosphorylation, SMAD2/3 form a complex with SMAD4 to regulate transcription.
LDLRAD4 interacts with SMAD2 and SMAD3, potentially competing with ZFYVE9 and SMAD4, thereby inhibiting intracellular signal propagation.
