This antibody detects endogenous levels of total Nesprin 1.
Target Information
Nesprin 1 and Nesprin 2 are members of a family of type II integral membrane proteins that are ubiquitously expressed. Tissue-specific alternative mRNA initiation and splicing generate multiple isoforms, with smaller forms truncated at the N-terminus. These proteins contain multiple clustered spectrin repeats, bipartite nuclear localization sequences, and a conserved C-terminal transmembrane domain.
Transient transfection studies with EGFP-fusion constructs have shown localization to the nuclear membrane, with the C-terminal TM domain essential for perinuclear localization. Nesprin 1 is developmentally regulated in smooth and skeletal muscle and relocates from the nuclear envelope to the nucleus and cytoplasm during myoblast differentiation. Nesprins may act as “dystrophins of the nucleus,” maintaining nuclear organization and structural integrity.
