Target Information

Crystallins are separated into two classes: taxon-specific (enzyme) and ubiquitous. The ubiquitous class forms the major proteins of the vertebrate eye lens, maintaining transparency and refractive index. Because lens fiber cells lose their nuclei during development, these proteins remain throughout life, making them extremely stable.

Mammalian crystallins are divided into alpha, beta, and gamma families; beta and gamma crystallins form a superfamily. Alpha and beta families are subdivided into acidic and basic groups. Crystallins have seven regions: four homologous motifs, a connecting peptide, and N- and C-terminal extensions. Gamma-crystallins are monomeric, symmetrical proteins lacking connecting peptides and terminal extensions, and are differentially regulated after early development.

Four gamma-crystallin genes (gamma-A through gamma-D) and three pseudogenes (gamma-E, gamma-F, gamma-G) are organized as a gene cluster. Aging or mutations in these genes are associated with cataract formation.
