Target Information
Cellular oxygen sensor that catalyzes, under normoxic conditions, the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins.
Hydroxylates specific prolines in the oxygen-dependent degradation domains (NODD and CODD) of HIF1A and HIF2A, targeting them for proteasomal degradation via the von Hippel-Lindau ubiquitination complex.
Under hypoxic conditions, hydroxylation is reduced, allowing HIFs to translocate to the nucleus and activate hypoxia-inducible genes.
EGLN3 (PHD3) plays a key role in limiting physiological activation of HIFs, regulating apoptosis in cardiomyocytes and neurons, and participating in DNA damage response via TELO2 hydroxylation.
