Target Information
Prolyl hydroxylase 4 (PHD4) is an enzyme that modifies HIF-alpha, playing a critical role in oxygen homeostasis. Unlike collagen-modifying prolyl hydroxylases found in the endoplasmic reticulum, HIF-related hydroxylases act intracellularly.
Under normoxic conditions, PHD4 hydroxylates specific prolyl residues in HIF-alpha, leading to its degradation via the von Hippel-Lindau ubiquitylation complex.
Under hypoxia, hydroxylation is reduced, stabilizing HIF-alpha and promoting angiogenesis. PHD4 functions as an oxygen sensor and catalyzes the posttranslational formation of 4-hydroxyproline in HIF-alpha proteins.
