Product Specific Information
M701 detects IFN gamma in Flow and Western blot applications, showing reactivity with human samples.
In sandwich ELISA, clone M700A (anti-IFN Gamma, clone 2G1) can be used as the coating antibody with M701B (biotinylated conjugate of clone B133.5) as the detection antibody.
Typical dilutions for sandwich ELISA:
- Coating: 1 µg/mL
- Detection: 0.125–0.25 µg/mL
The immunogen is recombinant human IFN gamma.
M701 detects IFN gamma with a predicted molecular weight of approximately 16 kDa.
Endotoxin level ≤ 10 EU/mg (tested by LAL assay).
Target Information
IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor produced mainly by T-lymphocytes and natural killer cells in response to antigens and cytokines.
It is a dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits, appearing as 25, 20, and minor 15.5 kDa bands on SDS-PAGE due to differential glycosylation.
Human IFN gamma does not cross-react with mouse IFN gamma.
Functions include:
- Antiviral and tumor antiproliferative activity
- Induction of MHC class I and II
- Macrophage activation
- Enhanced immunoglobulin secretion by B lymphocytes
IFN gamma acts via IFN gamma receptor I and II, activating the JAK-STAT pathway.
It shares ~40% sequence homology with mouse IFN gamma and is regulated by IL2, FGF basic, EGF (upregulation), and vitamin D3 or DMN (downregulation).
Mutations in the IFN gamma gene are associated with aplastic anemia.
