Description:
The JES3-9D7 monoclonal antibody reacts with human interleukin-10 (IL-10).

Applications Reported:
This antibody is suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for intracellular staining and flow cytometry using stimulated human peripheral blood cells with the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and BestProtocols® Protocol A.
Recommended usage: 5 µL (0.03 µg) per test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test (empirical).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with a 488 nm blue laser.
Recommended filter: 710/50 bandpass (or 695/40 alternative).
Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation; protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Excitation/Emission:
488 nm / 710 nm (Blue Laser)

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes and some lymphocytes. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages.
It promotes B cell survival, proliferation, and antibody production, and inhibits NF-kappa B activity.
IL-10 is involved in JAK-STAT signaling regulation.
Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis.
Associated diseases include Graft-Versus-Host Disease and HIV Type 1.
