Target Information

GAD-65 and GAD-67 are members of the group II decarboxylase family of proteins responsible for catalyzing the rate-limiting step in GABA (gamma-aminobutyric acid) production from L-glutamic acid.
Both proteins are found in the brain:

  • GAD-65: Localized to synaptic vesicle membranes in nerve terminals.
  • GAD-67: Distributed throughout the cell and responsible for basal GABA synthesis.

During increased neurotransmission demand, GAD-65 transiently activates to assist GABA production. Loss of GAD-65 impairs GABA neurotransmission, while loss of GAD-67 is lethal.
Two isoforms exist for GAD-67 due to alternative splicing: the predominant GAD-67 and minor GAD-25 forms.
