Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis including inflammatory prostaglandins. The conversion involves two reactions:
- Cyclooxygenase (COX) reaction – converts arachidonate to prostaglandin G2 (PGG2)
- Peroxidase reaction – reduces PGG2 to prostaglandin H2 (PGH2)
COX2 is constitutively expressed in tissues such as endothelium, kidney, and brain, and is up-regulated under pathological conditions like cancer and inflammation. Its up-regulation is associated with increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 is crucial for prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance. COX2 activity is naturally inhibited by calcitriol (active Vitamin D). Glucocorticoids repress PTGS2 gene activity by interfering with transcription initiation and elongation. COX2 is a known target of NSAIDs such as aspirin, which reduce inflammation-driven pain and swelling.
