Target Information
Myeloperoxidase (MPO) is a hemoprotein abundantly expressed in neutrophils and secreted during their activation. It exists as a covalently bound tetrameric complex of two glycosylated alpha chains (59–64 kDa) and two unglycosylated beta chains (~14 kDa), with a total molecular weight of ~150 kDa and a theoretical pI of 9.2.
MPO is a major target of anti-neutrophil cytoplasm antibodies (ANCA), which are markers for systemic vasculitis such as periarteriitis nodosa, microscopic polyarteriitis, and Churg-Strauss syndrome. Low to moderate levels of anti-MPO autoantibodies are also reported in rheumatoid arthritis. MPO is implicated in cardiovascular disease progression and possesses strong proinflammatory properties contributing to tissue injury. It plays a key role in the host defense system of polymorphonuclear leukocytes, catalyzing the production of hypohalous acids (primarily hypochlorous acid) and other toxic intermediates that enhance microbicidal activity.
